Persistence of the inuenza A/WSN/33 virus RNA at midbrain levels of immunodefective mice

چکیده
Strains of inuenza A virus are known to infect specic subpopulations of neurons in the mouse brain. Here we report that all segments of the genome of the neurotropic inuenza A virus, strain WSN/33, can persist in the brains of immunodefective transporter associated with Antigen Processing 1 (TAP1) mutant mice. Ten to 17 months after injection of virus into the olfactory bulbs, viral RNA encoding the nonstructural NS1 protein was detected in sections from the brain at midbrain levels by RT-PCR in almost all animals. Both negative-strand genomic RNA (vRNA) and positive-strand RNA, including mRNA, were found. RNA encoding nucleoprotein and polymerases, which form the replicative complex of the virus, were detected in fewer brains. RNA encoding envelope proteins were found only in occasional brains. No viral cDNA could be identied. This observation shows that certain regions of the brain in immunodefective mice may harbor the genome of inuenza A virus including the NS1 gene, the products of which may play a regulatory role in host-cell metabolism. Persistence of viral RNA in the brain of offspring to mice infected with inuenza A/WSN/33 virus during pregnancy
Epidemiological studies have indicated an association between inuenza A virus infections during fetal life and neuropsychiatric diseases. To study the potential for inuenza A virus infections to cause nervous system dysfunctions, we describe a mouse model using intranasal instillation of the mouse neuroadapted inuenza A/WSN/33 strain in pregnant mice. Viral RNA and nucleoproteinwere detected...

Infections caused by influenza A virus have been proposed to be associated with neuropsychiatric complications, the mechanisms of which remain to be unravelled. We here report that a neurotropic strain of influenza A virus (A/WSN/33) introduced into the olfactory bulbs of C57BL/6 (B6) mice, selectively attacks habenular, paraventricular thalamic, and brainstem monoaminergic neurons. In the habe...
متن کاملGlycosylation of neuraminidase determines the neurovirulence of influenza A/WSN/33 virus.
The neuraminidase (NA) gene of influenza A/WSN/33 (WSN) virus has previously been shown to be associated with neurovirulence in mice and growth in Madin-Darby bovine kidney (MDBK) cells. Nucleotide sequence analysis has indicated that the NA of WSN virus lacks a conserved glycosylation site at position 130 (corresponding to position 146 in the N2 subtype). To investigate the role of this carboh...
متن کاملVirions and intracellular nucleocapsids produced during mixed heterotypic influenza infection of MDCK cells.
Phenotypically mixed virus yields, obtained by coinfection of MDCK cells with influenza A/WSN/33 and B/Lee/40 viruses, contained both A/WSN/33 and B/Lee/40 NP proteins, as revealed by polyacrylamide gel electrophoresis of the purified 14C-amino acids-labeled virus. Virions were lysed with 0.6 M KCl-Triton X-100 buffer, and nucleocapsids were immunoprecipitated with antibodies against NP protein...
متن کاملInvestigation of Efficiency of Site-Specific Mutants of the Influenza Virus in Homological and Heterological Control Infection
The immunogenicity and protective efficacy of the site-specific mutants of A/WSN/33 (H1N1) strain of the influenza virus were compared with the similar characteristics of the cold-adapted (CA) reassortant obtained by crossing A/WSN/33 strain and CA A/ Krasnodar/101/35/59 (H2N2) strain of the influenza virus under homologous and heterologous control infection. The site-specific mutants had ts-mu...
